The Kinetics of the Decomposition of Peroxide by Catalase

چکیده

It has been shown that the experimental results obtained by Morgulis in a study of the decomposition of hydrogen peroxide by liver catalase at 20 degrees C. and in the presence of an excess of a relatively high concentration of peroxide are quantitatively accounted for by the following mechanisms. 1. The rate of formation of oxygen is independent of the peroxide concentration provided this is greater than about 0.10 M. 2. The rate of decomposition of the peroxide is proportional at any time to the concentration of catalase present. 3. The catalase undergoes spontaneous monomolecular decomposition during the reaction. This inactivation is independent of the concentration of catalase and inversely proportional to the original concentration of peroxide up to 0.4 M. In very high concentrations of peroxide the inactivation rate increases. 4. The following equation can be derived from the above assumptions and has been found to fit the experiments accurately. See PDF for Equation in which x is the amount of oxygen liberated at the time t, A is the total amount of oxygen liberated (not the total amount available), and K is the inactivation constant of the enzyme.

Preparation of Promoted Ni0.1Co0.9Fe2O4 Ferrite Nanoparticles and Investigation of Its Catalytic Activity on Decomposition of H2O2 and Optical Characterization of Pure Ni0.1Co0.9Fe2O4

Pure and ZnO-doped Ni0.1Co0.9Fe2O4 catalyst were prepared by co-precipitation method and thermal decomposition in air calcinated at 400-700°C and that treated with different amounts of zinc nitrate (0.46-2.25 w% ZnO). X-ray powder diffractometry, scanning electron microscopy (SEM) and BET analysis of nitrogen adsorption isotherms investigated the crystalline bulk structure and the surface area ...

Effect of pH upon the reaction kinetics of the enzyme-substrate compounds of catalase.

The existence of acid-linked groups in hemoproteins may be observed by studies of the effect of pH upon the nature of the enzyme and its compounds or upon the speed with which the enzyme compounds are formed. Since the activity of enzymatically active hemoproteins depends upon compound formation with the substrate, the effect of pH upon the overall activity may also demonstrate heme linkages. B...

Direct evidence for catalase as the predominant H2O2 -removing enzyme in human erythrocytes.

Decomposition of hydrogen peroxide (H2O2 ) at physiological levels was studied in human erythrocytes by means of a recently developed sensitive H2O2 assay. The exponential decay of H2O2 in the presence of purified erythrocyte catalase was followed down to 10(-9) mol/L H2O2 at pH 7.4. H2O2 decomposition by purified erythrocyte glutathione peroxidase (GPO) could be directly observed down to 10(-7...

The Decomposition of Hydrogen Peroxide by Liver Catalase

1. The velocity of decomposition of hydrogen peroxide by catalase as a function of (a) concentration of catalase, (b) concentration of hydrogen peroxide, (c) hydrogen ion concentration, (d) temperature has been studied in an attempt to correlate these variables as far as possible. It is concluded that the reaction involves primarily adsorption of hydrogen peroxide at the catalase surface. 2. Th...
